Will Zalatoris defeat Sepp Straka in a play-off in Memphis
The 25-year-old American, who has finished second in three majors, fired a four-under-par 66 to end on 15 under.
He started the day in third, one shot behind Straka, 29, who carded a 67.
Overnight leader JJ Spaun finished on five under, tied for 42nd, after a poor 78, while Cameron Smith finished nine under after a two-shot penalty.
Zalatoris was named PGA Tour rookie of the year after finishing runner-up at the Masters in 2021, and he matched that feat at the PGA Championship and US Open earlier this year.
He has risen to 14th in the world rankings after eight top-10 finishes on the PGA Tour, but that all-important Tour victory had so far eluded him.
The Texan was made to work for his win as Straka, who beat Ireland's Shane Lowry to claim his first Tour win at Palm Beach in February, pulled back a shot on Zalatoris in the back nine to force the play-off at TPC Southwind.
Eventually, the pair were separated on the third play-off hole after Straka could not match Zalatoris's bogey on the par-three 11th.
The St Jude Championship at TPC Southwind is the first of three tournaments to decide the Tour champion, with the winner picking up $18m (£14.8m).
Americans Brian Harman and Lucas Glover tied for third on 12 under, while seven players finished on 11 under, including England's Matthew Fitzpatrick and Spain's Jon Rahm.
